ABSTRACT

China is rich in antimony, boron, and vanadium mineral resources, which have been detected in environmental water bodies and drinking water. During the revision process of the "Standards for Drinking Water Quality (GB5749-2006)", research and evaluation are focused on three indicators: antimony, boron and vanadium. Vanadium is added and the limit value of boron is adjusted. This study reviews and discusses the technical contents related to the revision of the antimony, boron and vanadium, including the environmental presence levels, exposure status, health effects, and the revision of the standard limits of these three indicators. Suggestions are also made for the implementation of this standard.

ABSTRACT

Male reproductive infections are known to shape the immunological homeostasis of the testes, leading to male infertility. However, the specific pathogenesis of these changes remains poorly understood. Exosomes released in the inflammatory microenvironment are important in communication between the local microenvironment and recipient cells. Here, we aim to identify the immunomodulatory properties of inflammatory testes-derived exosomes (IT-exos) and explore their underlying mechanisms in orchitis. IT-exos were isolated using a uropathogenic Escherichia coli (UPEC)-induced orchitis model and confirmed that IT-exos promoted proinflammatory M1 activation with increasing expression of tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β), and interleukin-6 (IL-6) in vitro. We further used small RNA sequencing to identify the differential miRNA profiles in exosomes and primary testicular macrophages (TMs) from normal and UPEC-infected testes, respectively, and identified that miR-155-5p was highly enriched in IT-exos and TMs from inflammatory testes. Further study of bone marrow derived macrophages (BMDMs) transfected with miR-155-5p mimic showed that macrophages polarized to proinflammatory phenotype. In addition, the mice that were administrated IT-exos showed remarkable activation of TM1-like macrophages; however, IT-exos with silencing miR-155-5p showed a decrease in proinflammatory responses. Overall, we demonstrate that miR-155-5p delivered by IT-exos plays an important role in the activation of TM1 in UPEC-induced orchitis. Our study provides a new perspective on the immunological mechanisms underlying inflammation-related male infertility.

ABSTRACT

Objective To investigate the clinical symptoms experienced by patients with thoracic spinal tumors and verify the associated symptoms that are predictive of a decline in muscle strength in the lower limbs. Methods A single-center, retrospective cross-sectional study was conducted on in-patients diagnosed with epidural thoracic spinal tumors between January 2011 and May 2021. The study involved a review of electronic medical records and radiographs and the collection of clinical data. The differences in clinical manifestations between patients with constipation and those without constipation were analyzed. Binary logistic regression analyses were performed to identify risk factors associated with a decline in muscle strength in the lower limbs. Results A total of 227 patients were enrolled, including 131 patients with constipation and 96 without constipation. The constipation group had a significantly higher proportion of patients who experienced difficulty walking or paralysis compared to those without constipation prior to surgery (83.2% vs. 17.7%, χ2 = 99.035,P < 0.001). Constipation (OR = 9.522, 95%CI: 4.150-21.849, P < 0.001) and urinary retention (OR = 14.490, 95%CI: 4.543-46.213, P < 0.001) were independent risk factors for muscle strength decline in the lower limbs. Conclusions The study observed that patients with thoracic spinal tumors who experienced constipation symptoms had a higher incidence of lower limb weakness. Moreover, the analysis revealed that constipation and urinary retention were independent risk factors associated with a preoperative decline in muscle strength of lower limbs.

ABSTRACT

OBJECTIVE To establis h and validate a population pharmacokinetic model of docetaxel in malignant tumor patients. METHODS The clinical data of malignant tumor patients treated with chemotherapy regimen containing docetaxel in our hospital from June 2019 to December 2021 were retrospectively collected . According to the results of blood concentration detection , based on the three -compartment model the nonlinear mixed effect model (NONMEM)was used ;covariates(age, weight, height, body surface area ,Karnofsky performance scale ,total protein ,albumin, total bilirubin ,aspartate aminotransferase ,alanine aminotransferase and serum creatinine )affecting clearance (CL)were screened by “forward inclusion and backward exclusion ”; the population pharmacokinetic model of docetaxel was established . The model was tested for goodness -of-fit diagnosis and internal validation by Bootstrap . RESULTS A total of 264 measured blood concentrations of 132 patients with malignant tumors during chemotherapy were included . The covariates that had significant effect on CL of docetaxel were serum creatinine and total bilirubin (P<0.01). The results of Bootstrap analysis (parameter median values and 95% confidence intervals )were close to predict results of the established model ;the final model estimated that the population typical value of docetaxel CL was 37.82 L/h. CONCLUSIONS The population pharmacokinetic model of docetaxel in malignant tumor patients is established successfully , which can be used for the formulation and optimization of clinical individualized regimen .

ABSTRACT

Gut microbiota have been validated to play a pivotal role in metabolic regulation. As the most effective treatment for obesity and related comorbidities, bariatric surgery has been shown to result in significant alterations to the gut microbiota. Literature have recently suggested temporal and spatial features of alterations to the intestinal bacteria following bariatric surgery, which is possibly attributed to the gut adaptation to the surgical modification on the gastrointestinal tract. More importantly, the gut microbiota have been appreciated as a critical contributor to the metabolic improvements following bariatric surgery. Although not fully elucidated, the underlying mechanisms are associated with the molecular pathways mediating the crosstalk between gut microbiota and host . On the other hand, change of the gut microbiota has been found to be related to the prognosis of patients receiving bariatric surgery. Some studies even point out negative effects of the gut microbiota on certain surgical complications . In this review, we summarize the characteristics of alterations to the gut microbiota following bariatric surgery as well as its relevant impacts to better understand the role of gut microbiota in bariatric surgery.

ABSTRACT

The effect of radiotherapy on anti-tumor immunity is bidirectional, immunotherapy, especially the combination of immune checkpoint inhibitors (ICIs) and radiotherapy, can produce synergistic effects on anti-tumor immunity. Compared with conventional radiotherapy, stereotactic ablative body radiotherapy (SABR) can achieve high-precision and high-dose irradiation on target lesions, and has stronger anti-tumor immune activation effect. At the same time, due to the steep dose gradient, SABR can better protect the surrounding normal tissues, which is an effective means for the rapid control of local lesions in advanced non-small cell lung cancer (NSCLC). ICIs are an important component of standard treatment for advanced NSCLC. There is growing evidence that SABR in combination with ICIs can benefit patients with advanced NSCLC. This article reviews the biological basis and clinical research progress on the combination of these two therapies, aiming to provide reference for the domestic counterparts to better use this new treatment model.

ABSTRACT

ObjectiveTo study the possible molecular mechanism of baicalein (BAI)-mediated focal adhesion kinase (FAK) in the regulation of phosphatidylinositol 3-kinase/protein kinase B (PI3K/Akt) signaling pathway to inhibit the proliferation and migration of gastric cancer HGC-27 cells. MethodThe gastric epithelial GES-1 cells and gastric cancer HGC-27 cells were respectively treated with BAI (0, 5, 15, 25, and 50 μmol·L-1) for 48 h, and then methyl thiazolyl tetrazolium (MTT) assay was adopted to detect effect of BAI on cell proliferation. Western blot (WB) was employed to detect the expression of FAK and the proteins related to epithelial-mesenchymal transition (EMT) and PI3K signaling pathway after intervention with different concentrations of BAI. The HGC-27 cells stably overexpressing FAK were constructed with lentivirus-mediated transfection technique, and the transfection of FAK was detected through WB and green fluorescent protein (GFP). The cells were divided into empty vector (NC) group, BAI group, FAK overexpression group, and BAI-treated FAK overexpression group, and cell proliferation activity was detected by MTT assay. The colony formation and cell migration were observed via colony formation assay and Transwell migration assay, respectively. The expression of proteins involved in EMT and PI3K signaling pathways were detected by Western blot. ResultCompared with the NC group, BAI (15, 25 and 50 μmol·L-1) inhibited the proliferation of HGC-27 cells in a dose-dependent manner (P<0.05, P<0.01) while did not affect that of GES-1 cells. BAI (5, 15 and 25 μmol·L-1) down-regulated the expression level of p-FAK (P<0.05, P<0.01). Compared with NC group, FAK overexpression group showed up-regulated expression level of FAK in HGC-27 cells. The HGC-27 cells in both NC group and FAK overexpression group had green fluorescence. Compared with NC group, BAI inhibited the growth, colony formation, and migration, while FAK overexpression promoted those of HGC-27 cells. The treatment of FAK overexpression group with BAI inhibited the enhancement of cell proliferation and migration (P<0.05). WB showed that compared with NC group, BAI (15, 25 μmol·L-1) significantly up-regulated the expression of E-cadherin protein and down-regulated that of Vimentin, Snail, p-PI3K, and p-Akt protein in HGC-27 cells (P<0.05, P<0.01). Compared with NC group, FAK overexpression group showed down-regulated expression of E-cadherin, up-regulated expression of p-FAK, Vimentin, and Snail, and increased ratios of p-FAK/FAK, p-PI3K/PI3K and p-Akt/Akt (P<0.05). This phenomenon would be reversed after BAI treatment. ConclusionBAI can affect the proliferation and migration of gastric cancer HGC-27 cells by mediating FAK to regulate PI3K/Akt signaling pathway.

ABSTRACT

Ferroptosis is a form of iron-dependent regulated cell death. Evidence of its existence and the effects of its inhibitors on subarachnoid hemorrhage (SAH) is still lacking. In the present study, we found that liproxstatin-1 protected HT22 cells against hemin-induced injury by protecting mitochondrial functions and ameliorating lipid peroxidation. In in vivo experiments, we demonstrated the presence of characteristic shrunken mitochondria in ipsilateral cortical neurons after SAH. Moreover, liproxstatin-1 attenuated the neurological deficits and brain edema, reduced neuronal cell death, and restored the redox equilibrium after SAH. The inhibition of ferroptosis by liproxstatin-1 was associated with the preservation of glutathione peroxidase 4 and the downregulation of acyl-CoA synthetase long-chain family member 4 as well as cyclooxygenase 2. In addition, liproxstatin-1 decreased the activation of microglia and the release of IL-6, IL-1β, and TNF-α. These data enhance our understanding of cell death after SAH and shed light on future preclinical studies.

ABSTRACT

Sucrose is a natural product occurs widely in nature. In living organisms such as plants, sucrose phosphate synthase (SPS) is the key rate-limiting enzyme for sucrose synthesis. SPS catalyzes the synthesis of sucrose-6-phosphate, which is further hydrolyzed by sucrose phosphatase to form sucrose. Researches on SPS in recent decades have been focused on the determination of enzymatic activity of SPS, the identification of the inhibitors and activators of SPS, the covalent modification of SPS, the carbohydrate distribution in plants regulated by SPS, the mechanism for promoting plant growth by SPS, the sweetness of fruit controlled by SPS, and many others. A systematic review of these aspects as well as the crystal structure and catalytic mechanism of SPS are presented.

ABSTRACT

Climate change has been referred to as one of the greatest threats to human health, with reports citing likely increases in extreme meteorological events. In this study, we estimated the relationships between temperature and outpatients at a major hospital in Qingdao, China, during 2015-2017, and assessed the morbidity burden. The results showed that both low and high temperatures were associated with an increased risk of outpatient visits. High temperatures were responsible for more morbidity than low temperatures, with an attributed fraction (AF) of 16.86%. Most temperature-related burdens were attributed to moderate cold and hot temperatures, with AFs of 5.99% and 14.44%, respectively, with the young (0-17) and male showing greater susceptibility. The results suggest that governments should implement intervention measures to reduce the adverse effects of non-optimal temperatures on public health-especially in vulnerable groups.

ABSTRACT

Objective:To analyze the differentially expressed genes related to gastric cancer and analyze their related signaling pathways. Methods:The gastric cancer-related gene expression chips GSE63121 (miRNA) and GSE2685 (mRNA) were selected from the GEO database. After setting the screening criteria, the differentially expressed genes were obtained, and then the miRDB database was used to predict the common genes related to gastric cancer. GO and KEGG enrichment analyses were performed on consensus genes in the DAVID database. Genes with interactions were visualized using the STRING database to construct a PPI of differentially expressed genes. The relationship between differentially expressed genes and the survival rate of gastric cancer patients were predicted by the GEPIA database, and the meaningful differentially expressed genes were screened according to prognosis. Finally, the TIMER2.0 database was used to analyze the correlation between the finally screened genes and immune cell infiltration. Results:A total of 734 differentially expressed genes were obtained, of which 261 and 473 genes were up-regulated and down-regulated, respectively, and a total of 20 differentially expressed miRNA genes were obtained, of which 1 and 19 genes were up-regulated and down-regulated, respectively. Among these genes, a total of 103 genes were mapped, which were mainly involved in the regulation of immune system and metabolic process in gastric cancer. The screened significantly differentially expressed genes were CREB1 and ESR1, and gastric cancer patients had better prognosis with low expression of CREB1 and ESR1. The expressions of CREB1 and ESR1 were both positively correlated in gastric cancer tissue with the infiltration level of regulatory T cells (Tregs) in the tumor microenvironment (all P<0.05). Conclusions:CREB1 and ESR1 affect immune infiltration during the occurrence and development of gastric cancer. The high expression of CREB1 and ESR1 is associated with poor prognosis. The possible reason is that there are more Tregs cells in the tumor microenvironment. CREB1 and ESR1 are expected to become important targets for studying the pathogenesis and molecular mechanism of gastric cancer and its clinical application.

ABSTRACT

Objective:To study the anti-rheumatoid arthritis mechanism of Sanmiao Pill by using liquid chromatography-mass spectrometry. Methods:The rats were randomly divided into control group, model group and medicated group based on weight, 8 rats in each group. The model group and medicated group were treated by intradermal injection of 0.1 ml complete Freund's adjuvant on buttock(s). Rats of medicated group were given Sanmiao Pill solution 0.054 g/ml by gavage, 1 ml/100 g based on weight from the 7th day of experiment for 7 days. And then to score the degree of feet swelling of rats. After 48 hours of the last medication, to collecte the blood samples for metabolic fingerprint analysis. Then analized and processed the non-targeted contour mass spectrometry data (Continuum model) with unsupervised principal component method; identified the maximum metabolic differences between rheumatoid arthritis rats and healthy rats by dimension reduction technic. The differential ions were screened and locked by supervised pattern recognition analysis. Finally, with the help of the automated analysis software database like HMDB, identifiedthe structures and analized the correlated metabolic pathway. Results:Compared with the model group, the swelling degree of the feet (2.01 ± 0.19 ml vs. 2.27 ± 0.30 ml) in medicated group significantly decreased ( P<0.01). The HMDB data shows that the metabolic profiles of rats were mainly distributed in the control group and the model group, which proved that the rheumatoid arthritis rats treated with Sanmiao Pill had a pullback trend, and the overall metabolic level has a healthy state. With this analysis, 20 blood biomarkers related to rheumatoid arthritis were obtained, which involved the metabolic pathways of aminoacyl-tRNA biosynthesis, Glycine, Serine and Threonine metabolism, Alanine, Aspartic acid and Glutamic acid metabolism, Linoleic acid metabolism, Arachidonic acid metabolism. Conclusions:Sanmiao Pill has interventional effect on rheumatoid arthritis rats. The mechanism might be related with metabolic enzymes and metabolic pathways such as Alanine, Aspartic acid and Glutamic acid.

ABSTRACT

Objective:To investigate the mechanism of anti-proliferative scar of psoralen. Methods:Fibroblasts were cultured in vitro. The fibroblasts were divided into normal group (normal fibroblasts), scar group (hypertrophic scar fibroblasts), and TGF-β1 group (10 ng/ml TGF-β1 treated hypertrophic scar fibrils cells 5 min-12 hours), Smurf2 RNA interference group (Smurf2 siRNA transfected hypertrophic scar fibroblasts for 72 hours), psoralen group (10 μmol/L psoralen treatment of hypertrophic scar fibroblasts for 72 hours). The psoralen+TGF-β1 group (proliferative scar fibroblasts were added to the psoralen for 72 hours and then added to TGF-β1 for 6 hours). The expressions of Smurf2 and α-SMA protein were detected by Western blot method. The expression of type I collagen mRNA was detected by RT-PCR. The secretion of TGF-β1 protein was detected by ELISA.Results:Compared with the normal group, the expression of Smurf2 protein (0.83 ± 0.08 vs. 0.38 ± 0.07) in the scar group significantly increased ( P<0.05). Compared with the scar group, the expression of TGF-β1 (2.2 ± 0.18 vs. 4.2 ± 0.47) significantly decreased in Smurf2 RNA interference group ( P<0.05). The expression of Smurf2 (0.71 ± 0.06 vs. 0.42 ± 0.04) and α-SMA (1.42 ± 0.12 vs. 0.91 ± 0.09) proteinin the TGF-β1 group significantly increased ( P<0.05), and the expression of type I collagen mRNA (0.72 ± 0.09 vs. 0.41 ± 0.07) significantly increased ( P<0.05). The expression of Smurf2 (0.05 ± 0.01 vs. 0.42 ± 0.04) and α-SMA (0.71 ± 0.07 vs. 0.91 ± 0.09) proteinin in psoralen group significantly decreased ( P<0.05). The expression of collagen mRNA (0.12 ± 0.04 vs. 0.41 ± 0.07) significantly decreased ( P<0.05). Conclusions:Psoralen may inhibit the expression of α-SMA protein and decrease the expression of type I collagen by TGF-β1/Smurf2 signaling to inhibit scar formation.
